Germplasm resources refer to carriers that carry biological genetic information and have actual or potential use value. In order to keep the culture in a slow growth state for a long time, the subculture method is adopted when necessary, and the culture is regularly transferred to a fresh medium for re-cultivation. The method of cryopreservation is simple and easy, and does not require a large investment. It only needs an ordinary temperature-controllable light incubator or a small incubation room equipped with air-conditioning devices. After preservation, the plant material recovers quickly and has high activity, and it is extremely practical to preserve the original seed material. Special attention should be paid to the regular transfer for subculture before it can be stored for a longer period of time.

2. Inhibition of growth and preservation at room temperature. Inhibition of growth and preservation at room temperature refers to the addition of paclobutrazol, B9, CCC and other growth retardants or inhibitors to the preservation medium at room temperature to inhibit the growth rate of the preserved material, thereby prolonging preservation time. The advantages of this method are simple, easy to operate, and economical. It can also make the leaves of the test tube seedlings dark green, short and strong, and easy to root. The disadvantage is that it needs to be transferred regularly for subculture in order to store it for a longer time. In addition, long-term multiple subcultures may still induce mutations, produce genetic instability, and gradually reduce the redifferentiation ability of the materials.

3. Ultra-low temperature storage Ultra-low temperature storage is also called cryopreservation, which mainly refers to the state where cell metabolism and growth are basically stopped under the ultra-low temperature of liquid nitrogen (-196°C). Cells, tissues and organs will not cause changes in genetic traits under this condition, and can still preserve cell viability and morphogenesis. Provided with suitable conditions, planting resources can quickly restore the reproduction state, regenerate new plants and maintain the original genetic characteristics.
The use of ultra-low temperature preservation can not only preserve the germplasm of rare plants, but also preserve pollen, extend the life of pollen, and solve the difficulties in different flowering periods and hybridization of plants in different places.
The disadvantage of this method is that during the cooling and freezing process, if the water in the plant cells forms ice crystals and freezes, it will cause irreversible damage to the cell structure and cause death. Therefore, special attention should be paid to the handling procedures during the cryopreservation of materials.
